Operating Procedure:

  • Weigh 10.0g of sample under sterile conditions or use a pipette to aspirate 10.0ml for liquid samples, then transfer into a suitable sterile container (such as a sterile bag, dilution bottle, or Whirl-Pak bag).
  • Add 90.0ml of diluent to achieve a 1:10 ratio, then proceed with sample homogenization. Adjust pH if necessary. Commonly used diluents include Butterfield’s phosphate buffer, Maximum Recovery Diluent, or other appropriate solutions per BAM guidelines.
  • If necessary, continue with further dilution steps.
  • Open the Compact Dry plate cap, use a micropipette to take 1.0ml of the diluted sample and dispense it onto the center of the plate, then close the cap.
  • Invert the plate and incubate under specified conditions: 35 ± 2°C for 24 ± 2 hours according to AOAC; 37 ± 1°C for 24 ± 2 hours according to MicroVal and NordVal.

Reading results:

  • Select plates with a colony count in the range of 10–150 to ensure accuracy.
  • Blue or light blue colonies are the target objects for observation/identification.

Notes:

  • When the colony count exceeds 300 CFU, the entire plate surface may turn blue, making it difficult to distinguish individual colonies; in this case, sample dilution and retesting are required.
  • After reading results, plates must be sterilized by autoclaving (121°C for 30 minutes) before disposal.
