Operating procedure:

  • Aseptically weigh 10.0g of sample or use a pipette to take 10.0ml of liquid sample, then transfer into a suitable sterile container (e.g., sterile bag, dilution bottle, or Whirl-Pak bag).
  • Add 90.0ml of diluent to achieve a 1:10 ratio and homogenize the sample. Adjust pH if necessary. Commonly used diluents include Butterfield’s phosphate buffer, Maximum Recovery Diluent, or other appropriate solutions per BAM guidelines. If necessary, further dilution steps can be performed.
  • Open the Compact Dry plate cap, use a micropipette to take 1.0ml of the diluted sample and dispense it onto the center of the plate, then close the cap.
  • Invert the plate and incubate under standard conditions: AOAC International: 35 ± 2°C for 48 ± 3 hours; MicroVal & NordVal: 30 ± 1°C for 48 ± 3 hours.

Read results and count colonies:

  • Select plates with a colony count between 10–150 to ensure accuracy.
  • Observe red or colorless colonies.

Notes:

  • Some colonies may not display a clear red color.
  • If the colony count on the plate exceeds 300 CFU, the entire surface may turn red/pink, making it impossible to distinguish individual colonies; in this case, sample dilution is required.
  • After reading results, plates must be sterilized by autoclaving (121°C for 30 minutes) before disposal.
