Operating Procedure
1. Compact Dry Plate Preparation
- Open the aluminum storage pouch.
- Remove one set of 4 plates or the required number of plates (1–3 plates); keep the remainder in the sealed pouch to protect from light and humidity.
- Record all necessary information in the note section on the plate.
2. Sample Preparation

Diluent: Dissolve 7,650g NaCl, 0,724g Na₂HPO₄ và 0,210g KH₂PO₄ in 1L distilled water. Adjust pH to 7,4 using NaOH 1N. Sterilize by autoclaving at 121°C trong 15 minutes.

Sample Dilution:

Take 50g of sample, add 450ml of diluent → homogenize using a stomacher for 1 minute to obtain a 1:10 dilution.

Transfer 1ml of the 1:10 dilution into 9ml of diluent and mix thoroughly to obtain a 1:100 dilution.
3. Inoculation
- Use a pipette to aspirate 1ml of the diluted sample.
- Open the Compact Dry plate cap.
- Dispense 1ml of the sample onto the center of the plate surface.
- Close the cap.
4. Incubation
- Invert the capped plate and place it in the incubator.
- Incubate for 20-24 hours at 35°C..
Reading Results:
- Observe and count colonies from the underside of the plate.
- Select plates with a colony count between 10-150 to ensure accuracy.
- Blue or blue-green colonies are the target organisms for identification.
Notes:
- When the colony count exceeds 300 CFU, the entire plate surface may turn blue, making it impossible to distinguish individual colonies. In this case, sample dilution is required before retesting.
- After reading results, plates must be sterilized by autoclaving (121°C. for 30min) prior to disposal.
