Operating Procedure
- Sample Preparation: Aseptically weigh 10.0g of sample or pipette 10.0ml (for liquid samples) into a sterile container (e.g., sterile bag, dilution bottle, or Whirl-Pak bag).
- Dilution & Homogenization: Add 90.0ml of diluent to achieve a 1:10 dilution, then homogenize the sample. Adjust the pH if necessary. Commonly used diluents include Butterfield’s Phosphate-Buffered Diluent, Maximum Recovery Diluent (MRD), or other suitable solutions per BAM guidelines.
- Serial Dilution: If necessary, perform further decimal dilutions to reach the appropriate concentration.
- Inoculation: Open the lid of the Compact Dry plate. Using a micropipette, dispense 1.0ml of the diluted sample into the center of the plate. The sample will automatically diffuse across the surface. Replace the lid.
- Incubation: Invert the plates and incubate under the following specific conditions: MicroVal: 37 ± 1°C for 20–24 hours.; NordVal: 36 ± 1°C for 24 ± 2 hours.
Interpretation of Results
- Counting Range: Select plates containing 10–150 colonies for the most accurate count.
- Identification: Count all blue/blue-green colonies to determine the Enterococcus species count.
Notes
- High Concentration: If the bacterial load exceeds $10^4$ CFU per plate, individual colonies may not form, and the entire medium may turn blue. In such cases, further dilution of the sample is required.
- Interference: Note that Lactobacillus gasseri may produce false-positive results.
- Disposal: Following result interpretation, used plates must be sterilized via autoclaving (121°C for 30 minutes) before disposal.
